Grant given to Dr. Alfred L. George, Jr at the Northwestern University to determine if molecular mechanisms underlying ATP1A3-associated neurological disorders are amenable to antisense oligonucleotide therapeutic strategies. This research also aims to demonstrate the electrophysiological effects of antisense-oligonucleotide (ASO) directed knock down of ATP1A3 in human neurons.

The AHCF contributed to research at Brown University lead by Anne Hart that developed C. elegans AHC ATP1A3 models. Aim 1 of the project generated custom C. elegans lines using CRISPR-Cas9-based genome editing of the C. elegans ATP1A3, called eat-6, to create AHC models and controls for three patient alleles: D801N, E815K, and L839P. Aim 2 examined the consequences of inserting these patient missense alleles using multiple assessments of eat-6 function: hatch/growth, NMJ sensitivity to aldicarb, egg laying, and pharyngeal pumping rates. Here, provide a summary of results, along with a set of four Supplementary Files that contain additional analysis, figures and raw data.
