DURING THE FISCAL YEAR ENDING APRIL 2025, SOPHIE'S NEIGHBORHOOD FUNDED MAJOR RESEARCH INITIATIVES TO ADVANCE UNDERSTANDING AND TREATMENT OF MULTICENTRIC CARPO-TARSAL OSTEOLYSIS (MCTO) AN ULTRA RARE GENETIC BONE AND KIDNEY DISORDER. 1.ADVANCING MCTO DISEASE MECHANISMS THROUGH INNOVATIVE MOUSE MODELS SOPHIE'S NEIGHBORHOOD SUPPORTED RESEARCH AT HARVARD SCHOOL OF DENTAL MEDICINE INVESTIGATING BONE REMODELING ACTIVITY AND CELLULAR CHANGES MAFB MOUSE MODELS OF MCTO. THIS WORK INCLUDES TRANSCRIPTOMIC ANALYSES OF CELLS AND TISSUES WITHIN THE SKELETAL MICROENVIRONMENT TO UNCOVER MOLECULAR MECHANISMS DRIVING DISEASE PROGRESSION. 2.EXPANDING STEM CELL-BASED AND DEVELOPMENTAL MODELS OF MCTO IN COLLABORATION WITH BOSTON CHILDREN'S HOSPITAL, SOPHIE'S NEIGHBORHOOD CONTINUED TO FUND STUDIES ON MAFB AND DEVELOPMENTAL CHONDROGENESIS.
